Background
This product is a research-grade biosimilar of dargistotug, supplied as an unconjugated, non-therapeutic analog of the originator antibody built around its target, TIGIT. It is intended strictly for research use only and is not a clinical drug. Dargistotug is a fully human IgG1 (kappa) monoclonal antibody directed against the inhibitory immune checkpoint receptor TIGIT, developed as an immuno-oncology agent to relieve TIGIT-mediated suppression of T cells and NK cells. This biosimilar reproduces the antigen-binding specificity of the originator so that investigators can study TIGIT engagement, checkpoint blockade, and downstream immune effects in controlled in-vitro and functional settings without needing the proprietary clinical material. It is produced to research-grade specifications with low endotoxin (typically less than 1 EU/mg), and can be offered in bulk milligram-to-gram quantities to support assay development, screening cascades, and comparability work. As an unconjugated human IgG1 with a human target, it is well suited to binding, blocking, and effector-function studies and to serving as a reference or benchmark antibody alongside other anti-TIGIT reagents. It carries no clone designation, being defined instead by its originator-matched sequence and specificity.
TIGIT (T-cell immunoreceptor with Ig and ITIM domains) is a co-inhibitory receptor expressed on activated T cells, regulatory T cells, and NK cells. It binds the nectin-family ligands CD155 (PVR) and, with lower affinity, CD112, which are frequently upregulated on tumor cells and antigen-presenting cells. Engagement of TIGIT delivers inhibitory signals through its ITIM/ITT-like motifs and outcompetes the co-stimulatory receptor CD226 (DNAM-1) for the same ligands, dampening T-cell and NK-cell activation, proliferation, and cytokine output. TIGIT thereby contributes to immune exhaustion and tumor immune evasion. Blocking TIGIT is pursued to restore CD226-driven co-stimulation and enhance anti-tumor immunity, often in combination with PD-1/PD-L1 blockade, making it a prominent immune-checkpoint target in oncology research.
