Background
This product is a research-grade biosimilar of belrestotug (also known as EOS-448), an anti-TIGIT monoclonal antibody originated by iTeos Therapeutics and developed with GSK for immuno-oncology. It is an unconjugated, non-therapeutic analog supplied for research use only, built around the TIGIT target and matched to the originator's human IgG1, kappa framework. It is not the clinical drug product and is not intended for diagnostic or therapeutic use. Belrestotug is being evaluated clinically as a TIGIT antagonist, notably in combination with the anti-PD-1 antibody dostarlimab in non-small cell lung cancer. Unlike Fc-silent anti-TIGIT approaches, the originator retains a functional human IgG1 Fc, allowing engagement of Fc gamma receptors and antibody-dependent cellular cytotoxicity in addition to checkpoint blockade. This biosimilar is manufactured to research-grade standards with low endotoxin and is offered in bulk milligram to gram quantities, making it suitable as a reference or comparator reagent in binding, blockade, and effector-function studies. As with all ichor.bio biosimilars, it provides a cost-effective, well-characterised analog for in-vitro and functional research programs.
TIGIT (T-cell immunoreceptor with Ig and ITIM domains; also VSIG9 or VSTM3; UniProt Q495A1) is a co-inhibitory immune checkpoint receptor expressed on T cells and natural killer cells, with high expression on regulatory T cells within tumors. It is a 244-amino-acid type I membrane protein comprising an extracellular immunoglobulin variable domain, a transmembrane region, and a cytoplasmic tail bearing ITIM and ITT-like motifs that deliver inhibitory signals. TIGIT binds the nectin-family ligands CD155 (PVR) with high affinity and CD112 (PVRL2/nectin-2) with lower affinity, competing with the co-stimulatory receptor CD226 (DNAM-1). By outcompeting CD226 and signaling through its ITIM motifs, TIGIT dampens T-cell and NK-cell activation. Blocking TIGIT restores the CD226 co-stimulatory axis and reinvigorates anti-tumor immunity, which underlies its interest as a checkpoint target, frequently pursued alongside PD-1/PD-L1 blockade.
