Background
This product is an unconjugated, non-therapeutic recombinant analog of the anti-TIGIT antibody lancastotug, supplied for research use only. It reproduces the human IgG1/kappa framework and TIGIT-binding specificity of the originator molecule, allowing laboratories to study TIGIT blockade without sourcing clinical material. It is not the clinical drug and is not intended for human or veterinary use. The analog is useful as a defined, reproducible reference reagent for in-vitro and preclinical work: as a positive control in TIGIT binding and blocking assays, for benchmarking new anti-TIGIT candidates, for studying TIGIT/CD155 axis disruption on T and NK cells, and, given the native IgG1 backbone, for interrogating Fc-dependent effector functions such as ADCC and for antibody-drug-conjugate or bispecific development workflows. It is manufactured to research grade with low endotoxin (<1 EU/mg standard; ultra-low <0.5 EU/mg options) and is available in bulk milligram-to-gram quantities to support assay development, screening cascades, and reproducible lot-to-lot comparisons across extended research programs. TIGIT (T-cell immunoreceptor with Ig and ITIM domains; UniProt Q495A1) is a co-inhibitory receptor expressed on activated and regulatory T cells, follicular helper T cells, and natural killer cells. It binds the nectin-family ligands CD155 (PVR) and, with lower affinity, CD112 (PVRL2/nectin-2), competing with the co-stimulatory receptor CD226 (DNAM-1) for the same ligands. Because TIGIT binds CD155 with substantially higher affinity than CD226, it tips the balance toward inhibition. Ligand engagement drives ITIM/ITT-based intracellular signaling that dampens T-cell and NK-cell activation, cytokine production, and cytotoxicity. TIGIT is frequently co-expressed with PD-1 on exhausted tumor-infiltrating lymphocytes, making it a prominent immune-checkpoint target; blocking TIGIT aims to restore CD226-mediated co-stimulation and enhance anti-tumor immunity, often in combination with PD-1/PD-L1 blockade.
