Background
This product is an unconjugated, non-therapeutic recombinant analog of domvanalimab, a human IgG1-kappa antibody directed against TIGIT (T-cell immunoreceptor with Ig and ITIM domains; UniProt Q495A1). It is supplied strictly for research use only and is not the clinical drug, nor is it intended for human or veterinary use. Built around the same human TIGIT target as the originator, it is useful as a reference binder and tool reagent for characterising the TIGIT checkpoint axis in vitro and in preclinical settings. Typical research applications include use as a positive control or benchmarking standard in binding and blocking assays, epitope and competition studies, flow cytometry panels probing TIGIT-expressing lymphocytes, and as a comparator in ADCC or antibody-engineering workflows. The material is offered at research grade with low endotoxin (<1 EU/mg; ultra-low <0.5 EU/mg options), and is available in bulk milligram-to-gram quantities to support assay development, screening campaigns, and reproducible lot-to-lot comparisons. It allows laboratories to interrogate TIGIT biology without sourcing clinical-grade therapeutic material. TIGIT (T-cell immunoreceptor with Ig and ITIM domains) is a co-inhibitory receptor expressed on activated T cells, regulatory T cells, and natural killer (NK) cells. Its extracellular immunoglobulin domain binds the nectin-family ligands CD155 (PVR) and, with lower affinity, CD112 (nectin-2), competing with the co-stimulatory receptor CD226 (DNAM-1) for these ligands. Engagement of TIGIT delivers inhibitory signals through its cytoplasmic ITIM and ITT-like motifs, dampening T-cell and NK-cell activation, cytotoxicity, and cytokine production, while also reinforcing regulatory T-cell suppressive function. Because TIGIT restrains anti-tumour and antiviral immune responses and is frequently co-expressed with PD-1 on exhausted lymphocytes, it is a prominent immune-checkpoint target; blocking the TIGIT-CD155 interaction can shift the balance toward CD226-mediated co-stimulation and restored effector activity.
