Background
This product is an unconjugated, non-therapeutic recombinant analog of placulumab (also known as ART621, CEP-37247 or PN0621), an anti-TNF-alpha domain antibody construct. Unlike conventional monoclonal antibodies, the originator uses a bivalent domain antibody (dAb) format built from variable-domain binders fused to a human IgG Fc region, consistent with the [V-kappa]2 - Fc architecture shown on this card. The analog is engineered around the same human TNF-alpha target (UniProt P01375) and is intended strictly for research use, not for human or veterinary use. Supplied unconjugated, it is well suited to in-vitro and preclinical research applications: as a TNF-neutralisation reference, a positive/benchmark control alongside other anti-TNF biologics, a binding and epitope tool, and a scaffold for assay development or ADCC/effector characterisation. It is manufactured to research-grade low-endotoxin specifications (<1 EU/mg; ultra-low <0.5 EU/mg options), and is available in bulk milligram-to-gram quantities to support screening, standard-curve, and formulation work. It is a research analog, not the clinical drug. Tumour necrosis factor alpha (TNF-alpha, TNF, cachectin; gene TNF, UniProt P01375; TNF superfamily member TNFSF2) is a pleiotropic pro-inflammatory cytokine produced chiefly by activated macrophages and monocytes, as well as T cells, NK cells and other cell types. It is synthesised as a 26 kDa type II transmembrane protein that is cleaved by TACE/ADAM17 to release a 17 kDa soluble form; both membrane-bound and soluble TNF assemble into homotrimers. TNF signals through two receptors, TNFR1 (p55, TNFRSF1A) and TNFR2 (p75, TNFRSF1B), driving NF-kappaB and MAPK activation, inflammatory gene expression, and apoptotic or necroptotic cell death. It is a central mediator of chronic inflammatory and autoimmune disease, making it a validated therapeutic target for TNF-blocking biologics in conditions such as rheumatoid arthritis.
