Background
This product is an unconjugated, non-therapeutic recombinant research-grade analog of onercept, supplied for research use only. The clinical originator onercept is a recombinant human soluble p55 TNF receptor (TNFR1/TNFRSF1A) protein that acts as a decoy, binding free TNF and preventing it from engaging cell-surface receptors. It is therefore a soluble-receptor TNF antagonist rather than an anti-TNF monoclonal antibody, and this research analog carries no clone designation. Built around the TNF-binding activity of the originator, it is useful as a defined tool for probing TNF signalling in vitro: as a neutralizing/decoy reagent in ligand-binding and bioactivity assays, as a benchmarking control alongside other TNF antagonists (etanercept-like p75 constructs, adalimumab-like antibodies), and in preclinical characterization work. It is offered at low endotoxin (research grade less than 1 EU/mg; ultra-low less than 0.5 EU/mg) and in bulk milligram-to-gram quantities to support in-vitro and preclinical studies. It is not the clinical drug and is not for human or veterinary use. Tumor necrosis factor (TNF, cachectin; UniProt P01375) is a pro-inflammatory cytokine central to innate immunity, inflammation, and cell-death signalling. It is produced mainly by activated macrophages, monocytes, and T cells, first as a type II transmembrane protein that is cleaved by TACE/ADAM17 to release the soluble form; both membrane and soluble TNF assemble into homotrimers. TNF signals through two receptors: TNFR1 (p55, TNFRSF1A), broadly expressed and coupling to NF-kB, MAPK, and apoptotic/necroptotic pathways, and TNFR2 (p75, TNFRSF1B), more restricted to immune and endothelial cells. Downstream effects include upregulation of adhesion molecules, chemokine and cytokine induction, and regulation of cell survival versus death. Dysregulated TNF drives rheumatoid arthritis, inflammatory bowel disease, psoriasis, and other chronic inflammatory conditions, making it a validated target.
