Background
This is a research-grade, unconjugated non-therapeutic recombinant analog of the anti-CD47 antibody ligufalimab, engineered as a human IgG4-kappa built around the CD47 target (UniProt Q08722). It is supplied for research use only and is not the clinical drug, nor is it intended for human or veterinary use. As a functional binder to human CD47, it is useful as a tool reagent for probing the CD47-SIRPalpha "don't eat me" axis in vitro and in preclinical settings: mapping CD47 expression, blocking CD47-SIRPalpha engagement, and serving as a positive/reference control alongside other CD47-directed reagents or in cross-competition assays. The IgG4 framework, which limits Fc effector engagement relative to IgG1, makes it a useful backbone for dissecting blockade-driven versus Fc-driven phagocytosis. Provided at research grade with low endotoxin (<1 EU/mg; ultra-low <0.5 EU/mg options), it supports macrophage phagocytosis assays, binding/neutralization characterization, ADCC/ADC-development workflows, and biosimilar comparability studies. Bulk quantities (mg to gram scale) are available to support assay development, screening, and reproducible preclinical experiments.
CD47 (integrin-associated protein; UniProt Q08722) is a broadly expressed five-transmembrane cell-surface glycoprotein with a single extracellular IgV-like domain. Its principal counter-receptor is SIRPalpha on macrophages and other myeloid cells; CD47-SIRPalpha ligation delivers an inhibitory "don't eat me" signal that restrains macrophage phagocytosis. Many tumors upregulate CD47 to evade innate immune clearance, making the axis an oncology target. CD47 also binds thrombospondin-1, modulating nitric oxide signaling, vascular tone, and cell adhesion/migration, and it associates in cis with integrins to influence cell spreading and motility. Because CD47 is ubiquitously expressed, including on erythrocytes, target-mediated binding and potential red-cell effects are important considerations when designing and interpreting CD47-directed experiments.
