Background
This is an unconjugated, non-therapeutic recombinant analog of etanercept supplied strictly for research use. Etanercept is a soluble decoy built by fusing the extracellular ligand-binding portion of the p75 TNF receptor (TNFR2/TNFRSF1B) to the Fc region of human IgG1; it works by capturing soluble TNF alpha (and lymphotoxin-alpha) before they can engage cell-surface TNF receptors. The research-grade analog reproduces this TNF-binding decoy activity, making it a useful tool for interrogating TNF signalling in vitro: as a neutralising reagent to titrate or block TNF alpha in cell assays, as a positive/reference control alongside anti-TNF antibody biosimilars, and in binding, competition, and bioassay development work. Because it is provided unconjugated, at low endotoxin levels (research grade <1 EU/mg; ultra-low <0.5 EU/mg), and in bulk milligram-to-gram quantities, it suits reproducible, scaled preclinical and method-development experiments. It is not the clinical product and is not intended for human or veterinary use. Guidance provided for ichorbio to review. TNF alpha (tumour necrosis factor; gene TNF, UniProt P01375) is a pleiotropic pro-inflammatory cytokine of the TNF superfamily. It is produced chiefly by activated macrophages and monocytes, as well as T cells, NK cells and others, first as a type II transmembrane protein that is cleaved by the metalloprotease TACE/ADAM17 to release the soluble form; both forms assemble into biologically active homotrimers. TNF alpha signals through two receptors: TNFR1 (p55, TNFRSF1A), broadly expressed and able to drive NF-kB-dependent inflammatory gene expression as well as apoptotic and necroptotic cell death, and TNFR2 (p75, TNFRSF1B), more restricted and preferentially engaged by membrane TNF. Downstream pathways include NF-kB and MAPK activation, orchestrating inflammation, immune cell recruitment, and tissue remodelling. Dysregulated TNF alpha is central to rheumatoid arthritis, psoriasis, and inflammatory bowel disease, making it a validated therapeutic and research target.

Storage
Stable for at least one week when stored sterile at 2-8°C. For long term storage aseptically aliquot in working volumes without diluting and store at –80°C. Avoid Repeated Freeze Thaw Cycles.
