Background
This product is a research-grade biosimilar of afelimomab, supplied as an unconjugated, non-therapeutic analog of the originator molecule for research use only. Afelimomab is a murine-derived F(ab')2 antibody fragment directed against human tumor necrosis factor (TNF), and this preparation reproduces that target specificity and the F(ab')2, IgG3, kappa format described on the card. As a F(ab')2 fragment it retains bivalent antigen binding while lacking the Fc region, so it neutralizes soluble TNF without engaging Fc-mediated effector functions such as complement activation or Fc-receptor binding. It is not the clinical drug and is not intended for therapeutic or diagnostic use in humans or animals. The material is manufactured to research-grade endotoxin specifications (<1 EU/mg) and is available in bulk milligram-to-gram quantities, making it suitable as an in-vitro reference standard, positive control, or neutralizing reagent in TNF-focused assays. Because it targets human TNF, it is primarily used in cell-based and biochemical systems rather than routine mouse in-vivo dosing. Please confirm suitability for your specific application before use. Tumor necrosis factor (TNF, historically TNF-alpha; UniProt P01375) is a proinflammatory cytokine produced chiefly by activated macrophages and monocytes, as well as T cells, NK cells, and other lineages. It is synthesized as a type II transmembrane protein that is cleaved by the TACE/ADAM17 metalloprotease to release a soluble trimeric form. Both membrane-bound and soluble TNF signal through two receptors, TNFR1 (p55) and TNFR2 (p75), driving NF-kappaB and MAPK pathways that regulate inflammation, cell survival, and apoptosis. TNF is a central mediator in host defense but also a key driver of chronic inflammatory and autoimmune conditions, which is why TNF blockade is a well-established therapeutic strategy. Afelimomab binds soluble TNF and prevents its interaction with these receptors, making anti-TNF reagents valuable tools for studying cytokine signaling and neutralization.
