ICH5149 is a research-grade, unconjugated recombinant analog of the anti-IL-9 antibody enokizumab (originally MEDI-528), built on a human IgG1 kappa framework and designed to bind the human cytokine interleukin-9 (UniProt P15248). It is a non-therapeutic biosimilar intended strictly for research use only (RUO); it is not the clinical drug and is not for human or veterinary use. Because it reproduces the antigen-binding specificity of the originator against IL-9, it is well suited as a defined, reproducible reagent for target-engagement and cytokine-neutralization studies, as a positive/isotype-matched control in assay development, and as a building block for ADCC or antibody-drug-conjugate (ADC) development workflows. The product is supplied unconjugated so users can label, format, or conjugate it as needed. Low-endotoxin grades support sensitive cell-based work: research grade at less than 1 EU/mg and an ultra-low option at less than 0.5 EU/mg. Bulk quantities from milligram to gram scale are available to support screening, in-vitro pipelines, and preclinical standardization. No clone designation is implied. Interleukin-9 (IL-9; UniProt P15248) is a small secreted cytokine of the common gamma-chain family, produced most notably by Th9 cells and also by Th2, Th17, Treg, mast cells, ILC2s, and NKT cells. It signals through a heterodimeric receptor comprising the IL-9-specific alpha chain (IL9R) paired with the shared common gamma chain (IL2RG), driving JAK1/JAK3 activation and downstream STAT signaling (chiefly STAT5, with STAT1 and STAT3 contributions). IL-9 promotes mast cell expansion and activation, supports T-cell and mast-cell survival and proliferation, and stimulates airway epithelial mucus production. It is a key mediator in type 2/allergic inflammation and has been implicated in asthma, airway hyperresponsiveness and remodeling, atopic disease, and mucosal immunity, as well as in anti-tumor and anti-parasite responses. Its restricted, context-dependent expression makes IL-9 an informative node for studying Th9 biology and allergic inflammation.
