This product is a purified anti-mouse IFN gamma monoclonal antibody, clone XMG1.2, a rat IgG1 raised against mouse interferon gamma (UniProt P01580). XMG1.2 is one of the most widely used reference clones for detecting and neutralizing mouse IFN gamma, validated across ELISA capture, ELISPOT, intracellular flow cytometry, IHC on frozen sections, Western blot, CyTOF, and functional neutralization. The rat IgG1 isotype and its recognition of a neutralizing epitope make it well suited to in-vivo studies where the goal is to block IFN gamma bioactivity and interrogate its role in anti-tumour immunity, autoimmunity, infection, and inflammation. This ichor.bio preparation is manufactured to low endotoxin levels (research grade less than 1 EU/mg; ultra-low grade less than 0.5 EU/mg) and supplied in bulk milligram-to-gram quantities suitable for repeated dosing and multi-cohort animal cohorts. Interferon gamma (IFN gamma) is the sole type II interferon, a soluble cytokine produced predominantly by activated T cells (CD4 Th1 and CD8), natural killer cells, and NKT cells. It signals as a homodimer through the IFNGR1/IFNGR2 receptor complex, activating the JAK1/JAK2-STAT1 pathway to drive transcription of interferon-stimulated genes. IFN gamma is a master regulator of cell-mediated immunity: it activates macrophages and enhances their microbicidal and antigen-presenting capacity, upregulates MHC class I and class II expression, promotes Th1 differentiation, and orchestrates defence against intracellular pathogens. It also exerts anti-proliferative and immunomodulatory effects on tumour cells and can shape the tumour microenvironment. Dysregulated IFN gamma is implicated in autoimmune and inflammatory disease. Because it sits at the hub of Th1 responses, neutralizing IFN gamma in vivo is a common approach to dissect its contribution to immune and disease phenotypes.
