Interferon gamma (IFN gamma) is the sole type II interferon, a soluble cytokine produced predominantly by activated T cells (CD4 Th1 and CD8), natural killer cells, and NKT cells. It signals as a homodimer through the IFNGR1/IFNGR2 receptor complex, activating the JAK1/JAK2-STAT1 pathway to drive transcription of interferon-stimulated genes. IFN gamma is a master regulator of cell-mediated immunity: it activates macrophages and enhances their microbicidal and antigen-presenting capacity, upregulates MHC class I and class II expression, promotes Th1 differentiation, and orchestrates defence against intracellular pathogens. It also exerts anti-proliferative and immunomodulatory effects on tumour cells and can shape the tumour microenvironment. Dysregulated IFN gamma is implicated in autoimmune and inflammatory disease.
