Background
This product is an unconjugated, non-therapeutic recombinant analog of foralumab, a fully human IgG1 kappa monoclonal antibody directed against the CD3 epsilon (CD3E) chain of the T-cell receptor (TCR)-CD3 complex. It reproduces the antigen-binding specificity of the originator anti-CD3 molecule but is manufactured for research use only and is not intended for human or veterinary use. Because CD3-directed antibodies are central tools for studying T-cell activation, tolerance induction, and TCR signaling, a research-grade analog is useful as a defined, reproducible reagent for in-vitro assays and preclinical work: as a positive/comparator control in binding and cross-blocking experiments, in T-cell stimulation and modulation studies, and in bioassay development, antibody characterization, and assay standardization. It is offered at low endotoxin levels (research grade under 1 EU/mg; ultra-low under 0.5 EU/mg) and in bulk milligram-to-gram quantities to support screening, method development, and in-vitro standard curves. Being unconjugated, it can be used directly or labeled/formatted in-house for downstream applications. It is supplied as a characterization and control reagent for ichorbio to review, not as the clinical drug.
CD3 epsilon (CD3E, UniProt P07766) is an invariant subunit of the TCR-CD3 complex expressed on the surface of all mature T cells and thymocytes. Within the complex, CD3E pairs to form CD3D/CD3E (epsilon-delta) and CD3G/CD3E (epsilon-gamma) heterodimers, which assemble with the antigen-binding TCR alpha/beta heterodimer and the CD3 zeta (CD247) homodimer. CD3E carries immunoreceptor tyrosine-based activation motifs (ITAMs) in its cytoplasmic tail; when the TCR engages peptide-MHC, these ITAMs are phosphorylated by Src-family kinases LCK and FYN, recruiting ZAP-70 and initiating downstream signaling that drives T-cell activation, proliferation, and cytokine production. CD3E is essential for correct assembly, surface expression, and signaling of the receptor complex, and for normal thymic T-cell development. Its ubiquitous, T-cell-restricted expression makes CD3E a pan-T-cell marker and a key target for immunomodulation.
