Background
This product is a research-grade biosimilar of Dafsolimab, supplied as an unconjugated, non-therapeutic analog of the originator antibody for research use only. It is built around the Dafsolimab target, CD3 epsilon (CD3E), and is provided in a human IgG2b, kappa format. It is not the clinical drug and is not intended for therapeutic or diagnostic use in humans. The originator Dafsolimab is an anti-CD3E monoclonal antibody investigated for steroid-refractory acute graft-versus-host disease, including as part of combination approaches following hematopoietic stem cell transplantation; a distinct conjugated form (Dafsolimab setaritox, an antibody-drug conjugate) also exists but is a separate entity from this unconjugated biosimilar. As a research reagent, this material is intended to recapitulate the binding specificity of the originator so investigators can study target engagement and antibody function in controlled settings. It is manufactured to research-grade specifications with low endotoxin (typically less than 1 EU/mg), and is available in bulk milligram-to-gram quantities to support assay development, benchmarking, and mechanism-of-action studies. As with any biosimilar analog, users should confirm suitability and specificity for their particular application.
CD3 epsilon (CD3E; UniProt P07766) is a subunit of the T-cell receptor (TCR)-CD3 complex on the surface of T lymphocytes. Together with CD3-gamma, CD3-delta, and CD3-zeta chains and the TCR alpha/beta or gamma/delta heterodimers, it forms the multi-subunit complex that couples antigen recognition to intracellular signaling. Each CD3 chain carries immunoreceptor tyrosine-based activation motifs (ITAMs) in its cytoplasmic domain; upon TCR engagement, these are phosphorylated by Src-family kinases such as LCK and FYN, triggering downstream signaling and T-cell activation. Because of this central role, CD3E is a long-standing target for T-cell modulation and for redirecting or dampening T-cell responses. Antibodies against CD3E are widely used to study T-cell activation, immune-cell engagement, and tolerance-related mechanisms, making it a well-characterized target for functional immunology research.
