Background
This is a research-grade, unconjugated non-therapeutic recombinant analog of emirodatamab, an anti-CD3 arm-containing bispecific antibody built around the CD3 epsilon subunit of the T-cell receptor (TCR)/CD3 complex. It is supplied for research use only (RUO) and is explicitly not the clinical drug, not for human or veterinary use. The molecule is expressed as an asymmetric bispecific format (dual single-chain variable fragments paired with a single-chain Fc), reflecting the T-cell-engaging architecture in which one arm binds CD3 on T cells and the second arm binds a tumor-associated antigen to redirect T-cell cytotoxicity. As a research reagent it is useful as a positive control and benchmark in binding assays, for characterizing T-cell engager (TCE/BiTE-style) mechanism-of-action work, for comparative studies of CD3-directed constructs, and as a tool in in-vitro and preclinical cytotoxicity, T-cell activation, and cytokine-release model systems. It is offered at low endotoxin (research grade under 1 EU/mg; ultra-low grade under 0.5 EU/mg), and available in bulk milligram-to-gram quantities to support assay development and scale-up. CD3 epsilon (CD3E, UniProt P07766) is an invariant signaling subunit of the T-cell receptor (TCR)/CD3 complex expressed on all mature T lymphocytes. Within the complex, CD3 epsilon associates non-covalently with CD3 gamma and CD3 delta to form heterodimers, alongside the CD3 zeta homodimer, clustering around the antigen-recognizing TCR alpha/beta chains. CD3 epsilon carries an immunoreceptor tyrosine-based activation motif (ITAM) in its cytoplasmic tail; upon TCR engagement of peptide-MHC, Lck-mediated phosphorylation of these ITAMs recruits ZAP-70 and initiates the downstream signaling cascade driving T-cell activation, proliferation, and effector function. Because CD3 epsilon is broadly and stably surface-expressed and functionally central to TCR signaling, it is the canonical target for the T-cell-engaging arm of bispecific antibodies and BiTE-style constructs, which crosslink CD3 epsilon to a tumor antigen to trigger polyclonal, MHC-independent redirected cytotoxicity.
