Background
This is a research-grade, unconjugated recombinant analog of natalizumab, a human IgG4 kappa antibody directed against the alpha-4 integrin subunit (CD49d, ITGA4). It reproduces the antigen-binding specificity of the originator molecule but is manufactured strictly for research use only and is not the clinical therapeutic; it is not intended for human or veterinary use. Supplied unconjugated, it is well suited to in-vitro and preclinical work such as blocking and neutralisation assays, competition and epitope-binning studies, flow cytometry and cell-adhesion experiments, and as an isotype-matched or positive-control reagent when characterising anti-CD49d reagents. Because it carries the same variable-region specificity as the reference drug, it is a convenient benchmark for biosimilarity and comparability studies and for developing binding or potency assays. Available at low endotoxin (research grade below 1 EU/mg; ultra-low grade below 0.5 EU/mg) and in bulk milligram-to-gram quantities, it supports assay development, screening campaigns, and preclinical model work requiring consistent lot-to-lot performance.
CD49d is the alpha-4 integrin subunit (ITGA4, UniProt P13612). It pairs with the beta-1 subunit (CD29) to form VLA-4 (alpha4beta1) and with the beta-7 subunit to form the alpha4beta7 integrin. VLA-4 binds vascular cell adhesion molecule-1 (VCAM-1) and the CS-1 domain of fibronectin, mediating firm adhesion and transendothelial migration of lymphocytes, monocytes, and other leukocytes across activated endothelium. Alpha4beta7 binds MAdCAM-1 and directs lymphocyte homing to gut-associated mucosal tissue. Through these interactions the alpha-4 integrins govern leukocyte trafficking into inflamed tissue and the central nervous system, and contribute to immune cell activation and retention. Blocking CD49d interrupts leukocyte adhesion and extravasation, which is the rationale underlying alpha-4-directed antibodies in inflammatory and autoimmune research contexts.
