Background
This product is a research-grade biosimilar of vonsetamig, supplied as an unconjugated, non-therapeutic analog of the originator molecule for research use only. It is not the clinical drug and is not intended for diagnostic or therapeutic use. Vonsetamig (also referenced as REGN5459) is a BCMAxCD3 bispecific T-cell engaging antibody developed for relapsed or refractory multiple myeloma. It is built to bridge B-cell maturation antigen (BCMA/TNFRSF17), which is highly expressed on malignant plasma cells, with the CD3 epsilon subunit of the T-cell receptor complex on T cells. By co-engaging both antigens, the molecule redirects and activates polyclonal T cells against BCMA-positive target cells independently of TCR specificity, driving target-cell lysis. This biosimilar analog reproduces the dual-target binding architecture around these two antigens and is intended to support in-vitro and functional research such as binding characterization, T-cell redirection and cytotoxicity assays, cytokine-release readouts, and as a reference or control reagent. It is manufactured to a research grade with low endotoxin and is available in bulk milligram to gram quantities. This molecule engages two antigens. TNFRSF17 (BCMA) is a TNF-receptor-superfamily member expressed largely restricted to the plasma-cell lineage and late memory B cells; it binds APRIL and BAFF to promote plasma-cell survival and is strongly upregulated on malignant plasma cells in multiple myeloma, making it an attractive tumor-associated target. CD3E encodes the CD3 epsilon chain, an invariant signaling subunit of the CD3/T-cell receptor complex present on all mature T cells; clustering of CD3 transduces activating signals that trigger T-cell proliferation, cytokine secretion, and cytotoxic effector function. A BCMAxCD3 bispecific simultaneously binds BCMA on tumor cells and CD3 epsilon on T cells, forming an immunological synapse that redirects T-cell cytotoxicity toward BCMA-expressing cells regardless of native antigen specificity, with concomitant T-cell activation and cytokine release.
