Divozilimab Biosimilar (ICH5483) is an unconjugated, non-therapeutic research-grade analog of the originator anti-CD20 antibody divozilimab, provided for research use only and not for clinical or diagnostic application. The originator is a humanized, afucosylated IgG1 monoclonal antibody directed against human CD20 (MS4A1), developed as a B-cell-depleting agent and studied in relapsing multiple sclerosis. This biosimilar is engineered around the same molecular target, CD20, and is intended as a functional tool for in-vitro and comparator studies rather than as a substitute for the approved drug. It is offered in the human IgG1, kappa format matching the originator isotype. As a research reagent it is well suited to target-binding characterization, comparator and control experiments, and mechanistic assays exploring CD20 engagement and antibody effector function. The material is supplied as a purified antibody at research-grade endotoxin levels, in bulk milligram-to-gram quantities suitable for demanding experimental workflows. It supports investigators who require a consistent, cost-effective analog of a clinically studied anti-CD20 antibody for benchmarking, assay development, and translational research programs, without the sourcing constraints of clinical drug product. CD20, encoded by MS4A1, is a non-glycosylated four-transmembrane surface phosphoprotein of the MS4A family expressed on B lymphocytes from the pre-B through mature and memory stages, but absent on early pro-B cells and terminally differentiated plasma cells. It functions as a store-operated calcium channel component and contributes to B-cell activation, proliferation, and differentiation. Because it is stably and selectively expressed across most of the B-cell lineage and is not internalized or shed appreciably upon antibody binding, CD20 is a well-validated target for antibody-mediated B-cell depletion. Anti-CD20 antibodies eliminate B cells through antibody-dependent cell-mediated cytotoxicity, complement-dependent cytotoxicity, and phagocytosis; afucosylation enhances FcgammaRIIIa engagement and ADCC potency. This depletion mechanism underlies the therapeutic rationale for anti-CD20 agents in B-cell malignancies and autoimmune diseases such as multiple sclerosis.
