Background
This product is an unconjugated, non-therapeutic recombinant analog of ficlatuzumab, provided for research use only. Ficlatuzumab is a humanized IgG1 kappa antibody directed against human hepatocyte growth factor (HGF), the ligand for the MET receptor tyrosine kinase. By binding HGF, the analog neutralizes ligand-driven activation of MET signaling, making it a useful tool for interrogating the HGF/MET axis in vitro and in preclinical models. Because it reproduces the binding specificity of the originator without being formulated or intended for clinical use, it serves well as a positive control, a benchmarking reagent for assay development, a neutralizing antibody in cell-based signaling studies, and a starting point for ADCC or antibody-drug-conjugate development workflows. It is supplied as bulk material at milligram-to-gram scale with low endotoxin (research grade less than 1 EU/mg; ultra-low grade less than 0.5 EU/mg), suitable for demanding functional assays. It is not the clinical drug and is not for human or veterinary use. Hepatocyte growth factor (HGF, UniProt P14210), also called scatter factor, is a multidomain glycoprotein secreted mainly by mesenchymal and stromal cells. It is produced as an inactive single-chain precursor that is proteolytically cleaved into a disulfide-linked alpha/beta heterodimer to become active. HGF is the sole high-affinity ligand for the receptor tyrosine kinase MET (c-Met). Ligand binding drives MET dimerization, autophosphorylation, and downstream signaling through PI3K/AKT, RAS/MAPK, and STAT3 pathways, promoting cell proliferation, survival, motility (scattering), morphogenesis, and angiogenesis. Physiologically the HGF/MET axis is central to tissue regeneration, wound healing, and embryonic development. Aberrant HGF/MET signaling contributes to tumor growth, invasion, metastasis, and resistance to targeted therapies, which makes HGF neutralization an actively studied intervention point in oncology research.
