Background
This product is an unconjugated, non-therapeutic recombinant analog of inotuzumab, supplied for research use only. It is built around CD22 (Siglec-2), the B-cell surface antigen that the originator antibody engages, and reproduces the human IgG4-kappa framework so investigators can study antigen recognition and antibody behaviour in defined systems. It is not the clinical drug and, importantly, is not conjugated to a payload; the marketed originator is delivered as an antibody-drug conjugate, so this bare analog is intended as a research tool and a starting point for conjugation or ADC-development chemistry rather than as a therapeutic. Typical applications include CD22-binding and specificity assays, use as an isotype-matched or target-specific reference standard, flow-cytometry and imaging staining of CD22-positive cells, and preclinical characterisation work. Material is offered at low endotoxin (research grade less than 1 EU/mg; ultra-low grade less than 0.5 EU/mg) and in bulk milligram-to-gram quantities to support assay development, screening campaigns and reproducible in-vitro studies. Not for human or veterinary use. CD22 (Siglec-2) is a type I transmembrane sialic-acid-binding immunoglobulin-like lectin restricted almost entirely to the B-cell lineage. Its extracellular region carries multiple Ig-like domains, with the N-terminal V-set domain recognising alpha-2,6-linked sialic acid on glycoproteins, mediating both homotypic and heterotypic interactions. CD22 functions as an inhibitory co-receptor of the B-cell receptor: antigen engagement drives phosphorylation of cytoplasmic immunoreceptor tyrosine-based inhibitory motifs (ITIMs), which recruit the phosphatase SHP-1 to dampen BCR signalling and set B-cell activation thresholds. It also contributes to B-cell survival, adhesion and homing to bone marrow. Because CD22 is expressed across most B-cell malignancies yet absent from stem cells, and because it internalises rapidly on antibody binding, it is an established target for antibody and antibody-drug-conjugate approaches in B-cell disease.
