Background
This product is an unconjugated, non-therapeutic recombinant analog of firastotug, a fully human IgG1-kappa monoclonal antibody directed against human cytotoxic T-lymphocyte-associated protein 4 (CTLA-4, UniProt P16410). It is supplied for research use only and is not the clinical drug; it is not intended for human or veterinary use. Built around the CTLA-4 target, the antibody is useful as a well-characterized reference reagent for benchmarking anti-CTLA-4 binding and blocking activity, as a positive control in checkpoint-blockade assays, and as a comparator in immuno-oncology screening workflows. It supports in-vitro and preclinical studies of T-cell co-stimulation, regulatory T-cell biology, and antibody-dependent effector mechanisms, and can serve as a tool for ADCC/ADC-development and Fc-engineering investigations. The antibody is manufactured to research grade with low endotoxin (<1 EU/mg research grade; <0.5 EU/mg ultra-low endotoxin options) and is available in bulk quantities from milligram to gram scale, making it suitable for high-throughput assay development, in-vitro standards, and reproducible batch-to-batch comparisons.
CTLA-4 (CD152) is an inhibitory immune checkpoint receptor expressed by activated conventional T cells and constitutively by regulatory T cells (Tregs). It is a close homolog of the co-stimulatory receptor CD28 and competes with CD28 for the shared B7 ligands CD80 (B7-1) and CD86 (B7-2) on antigen-presenting cells, binding them with substantially higher avidity. By outcompeting CD28 and physically removing B7 ligands from APC surfaces via trans-endocytosis, CTLA-4 dampens the co-stimulatory "second signal" required for full T-cell activation, thereby raising the threshold for T-cell priming and enforcing peripheral tolerance. Treg-expressed CTLA-4 is central to their suppressive function. Loss of CTLA-4 causes lethal lymphoproliferation in mice, underscoring its role as a non-redundant negative regulator. Antibody blockade of CTLA-4 releases this brake, enhancing T-cell priming and anti-tumor immunity.
