Clone DATK32 is a rat IgG2a monoclonal antibody raised against mouse LPAM1, the integrin alpha4beta7 (also called integrin alpha-4/beta-7 heterodimer). It recognizes the combinatorial alpha4beta7 epitope rather than the individual alpha4 or beta7 subunits, making it a specific reporter and functional modulator of the intact gut-homing integrin. This lot is offered as a low-endotoxin, RUO-grade preparation suited to in-vivo and functional work: it is validated for Western Blot and blocking applications, and its capacity to interfere with alpha4beta7-dependent adhesion makes it a common tool for interrogating lymphocyte trafficking to intestinal tissue. The rat IgG2a format on a mouse target allows straightforward pairing with appropriate isotype controls and secondary reagents. Low endotoxin (research grade <1 EU/mg; ultra-low <0.5 EU/mg) reduces the risk of confounding innate-immune activation in animal experiments, and bulk milligram-to-gram scale supports multi-cohort in-vivo dosing studies. LPAM1 (lymphocyte Peyer's patch adhesion molecule 1) is the integrin alpha4beta7 heterodimer, formed by the alpha4 (ITGA4/CD49d) and beta7 (ITGB7) subunits. It is expressed on subsets of lymphocytes, including memory/effector T cells, B cells, and innate lymphoid cells, and serves as the principal gut-homing receptor. Its main counter-receptor is MAdCAM-1 (mucosal addressin cell adhesion molecule-1), which is displayed on high endothelial venules of Peyer's patches and mesenteric lymph nodes and on lamina propria venules. Alpha4beta7 also binds VCAM-1 and fibronectin with lower avidity. Engagement of MAdCAM-1 mediates rolling and firm arrest of circulating lymphocytes, directing their entry into gut-associated lymphoid tissue and intestinal mucosa. This axis is central to mucosal immune surveillance and to the pathogenesis of inflammatory bowel disease, making alpha4beta7 a validated therapeutic target for blockade.
