Effector-silenced ADC formats require non-targeting controls that share the same Fc engineering. Bearing LALA-PG mutations, this human IgG1 kappa isotype control is directed against hen egg lysozyme, a non-mammalian antigen, and therefore provides no specific binding while acting as a matched negative reference. It is conjugated with MMAE via a PEG8 linker at a drug-to-antibody ratio of 4 (DAR4), allowing researchers to benchmark payload and Fc-silenced framework effects independent of antigen binding when setting background and gating. The reagent is research use only, supplied in a low-endotoxin format suitable for in vivo use and produced in scalable milligram (mg) to gram (g) quantities for bulk research.
