Background
This is an unconjugated, non-therapeutic recombinant analog of ranibizumab, an anti-VEGF-A biologic, supplied strictly for research use only. It is built to recognise human vascular endothelial growth factor A (VEGF-A, UniProt P15692) and reproduces the target-binding behaviour of the originator so investigators can use it as a defined reference reagent rather than as a clinical product. Note that clinical ranibizumab is an antigen-binding fragment (Fab) rather than a full antibody; this research-grade analog is offered as a human IgG1 kappa format for laboratory work, so users studying Fab-specific properties should confirm the format suits their assay. Typical applications include VEGF-A neutralisation and binding studies, isotype/positive controls, standard curves, benchmarking of biosimilar or novel anti-VEGF candidates, and preclinical in-vitro angiogenesis models. It is manufactured at low endotoxin (research grade <1 EU/mg; ultra-low <0.5 EU/mg options) and available in bulk milligram-to-gram quantities to support screening and scale-up. It is not the clinical drug and is not for human or veterinary use. VEGF-A (UniProt P15692) is the prototypic member of the vascular endothelial growth factor family and the principal driver of physiological and pathological angiogenesis. It is a secreted, disulphide-linked homodimeric glycoprotein produced as several splice isoforms (for example VEGF121, VEGF165, VEGF189) that differ in heparin and neuropilin binding. VEGF-A signals mainly through the receptor tyrosine kinases VEGFR-1 (FLT1) and VEGFR-2 (KDR), with VEGFR-2 mediating most pro-angiogenic effects, promoting endothelial proliferation, migration, survival and vascular permeability. Hypoxia, via HIF-1alpha, is a key inducer of VEGF-A expression. Dysregulated VEGF-A underlies neovascular (wet) age-related macular degeneration, diabetic macular oedema, retinal vein occlusion and tumour angiogenesis, which is why VEGF-A neutralisation is a central therapeutic and research strategy.
