Background
This product is an unconjugated, non-therapeutic recombinant analog of lonigutamab, a humanized anti-IGF-1R IgG1 (kappa) antibody, supplied strictly for research use. It is built around the IGF-1R (insulin-like growth factor 1 receptor) target and is intended to reproduce the antigen-binding behavior of the originator molecule so investigators can study IGF-1R engagement, receptor internalization/degradation, and downstream signaling in controlled experiments. It is not the clinical drug and is not intended for human or veterinary use. The analog is useful as a positive binding control, as a reference reagent in receptor-blockade and neutralization assays, for ADCC/effector-function and antibody-engineering workflows, and in preclinical in-vitro and cell-based models where a defined anti-IGF-1R IgG1 tool is needed. It is offered as low-endotoxin research grade (<1 EU/mg), with ultra-low-endotoxin (<0.5 EU/mg) and bulk milligram-to-gram formats available to support in-vitro standards, assay development, and scale-up. Because IGF-1R is a human target, the reagent is primarily suited to human-cell and biochemical systems rather than routine rodent in-vivo dosing.
IGF-1R (IGF1R, CD221; UniProt P08069) is a transmembrane receptor tyrosine kinase in the insulin-receptor family. It is synthesized as a proreceptor that is cleaved into alpha and beta chains assembled as a disulfide-linked (alpha-beta)2 heterotetramer. Ligand binding by IGF-1 (and, with lower affinity, IGF-2 and insulin) triggers beta-chain autophosphorylation and activation of the PI3K-AKT and RAS-MAPK pathways, driving cell proliferation, growth, survival, and differentiation. IGF-1R shares high homology with the insulin receptor (INSR; UniProt P06213) and can form IGF-1R/INSR hybrid receptors, which is why both accessions appear on this card; the validated therapeutic target of the originator, however, is IGF-1R. In thyroid eye disease, IGF-1R signaling in orbital fibroblasts promotes hyaluronan production and inflammation, providing the rationale for anti-IGF-1R blockade.
