Background
This product is a research-grade biosimilar of Dalotuzumab, supplied as an unconjugated, non-therapeutic analog of the originator antibody for research use only. It is built around the type 1 insulin-like growth factor receptor (IGF1R) and is intended as a laboratory research reagent, not as a clinical drug. Dalotuzumab (also known as MK-0646) is a humanized IgG1, kappa antibody that binds IGF1R and blocks receptor activation by its ligands. This analog reproduces the target specificity of the originator so that investigators can study IGF1R biology, ligand-blocking activity, and antibody-based approaches to the IGF axis in controlled in-vitro and functional assays. As with all ichor.bio research antibodies, it is manufactured to a low-endotoxin specification (research grade less than 1 EU/mg) and is available at scalable bulk quantities from milligram to gram scale to support reproducible, larger studies. It carries an in-vivo research-grade positioning, though because IGF1R is a human target most applications are functional and cell-based rather than mouse dosing studies. It is offered strictly for research use only. IGF1R (type 1 insulin-like growth factor receptor) is a transmembrane receptor tyrosine kinase and the principal receptor for the growth factors IGF-1 and IGF-2. It is a disulfide-linked alpha2-beta2 heterotetramer: ligand binding to the extracellular alpha subunits triggers autophosphorylation of the intracellular beta-subunit kinase domains, recruiting adaptors such as IRS-1 and Shc and activating the PI3K/AKT and RAS/MAPK pathways that drive cell proliferation, survival, and growth. IGF1R is closely related to the insulin receptor (INSR) and can form IGF1R/INSR hybrid receptors, which is relevant to signaling crosstalk and to interpreting target-specificity experiments. Because sustained IGF1R signaling supports tumor cell growth and can mediate resistance to other therapies, the receptor is a long-standing subject of oncology research, making reagents that engage and block IGF1R useful tools for dissecting this pathway.
