Background
This product is an unconjugated, non-therapeutic recombinant analog of cixutumumab, a fully human IgG1 kappa antibody directed against the type 1 insulin-like growth factor receptor (IGF1R). It is manufactured for research use only and is not the clinical drug; it is not intended for human or veterinary use. Built around the IGF1R target, it reproduces the binding specificity of the originator antibody, making it a useful reference reagent for in-vitro and preclinical work: as a positive control in IGF1R binding and blockade assays, a benchmarking tool against other anti-IGF1R antibodies, a capture or detection reagent, and a starting material for ADCC characterization or antibody-drug-conjugate development workflows. Supplied at research-grade low endotoxin (<1 EU/mg), with an ultra-low endotoxin option (<0.5 EU/mg) suitable for sensitive cell-based and in-vivo standard applications, and available in bulk milligram-to-gram quantities to support assay development, screening campaigns, and scale-up. The human IgG1 kappa framework allows it to serve as an isotype-matched effector-competent comparator when studying Fc-mediated function against IGF1R-expressing cells.
IGF1R (insulin-like growth factor 1 receptor; UniProt P08069) is a disulfide-linked (alpha-beta)2 transmembrane receptor tyrosine kinase. Ligand binding by IGF-1 or IGF-2 to the extracellular alpha subunits triggers autophosphorylation of the intracellular beta-subunit kinase domains, recruiting IRS-1/2 and Shc adaptors and activating the PI3K-AKT and RAS-MAPK cascades that drive cell proliferation, survival, growth, and resistance to apoptosis. IGF1R is central to normal somatic growth and is frequently implicated in tumor biology, where signaling supports transformation and treatment resistance. It is structurally homologous to the insulin receptor (INSR; UniProt P06213, the second accession listed) and can form IGF1R/INSR hybrid receptors, a source of cross-talk and therapeutic-escape considerations. Cixutumumab binds IGF1R selectively, blocking ligand engagement and promoting receptor internalization and downregulation.
