Background
This product is an unconjugated, non-therapeutic recombinant analog of blosozumab, an anti-sclerostin (SOST) antibody, supplied for research use only. It is built around blosozumab's target, human sclerostin, and is provided as a full-length human IgG4 (S228P stabilized hinge; C-terminal Lys removed) kappa immunoglobulin. It is not the clinical drug and is not intended for human or veterinary use. The format is well suited to bench research: as a positive/reference control in sclerostin binding and neutralization assays, for benchmarking or characterizing other anti-SOST antibodies, for epitope-competition and pharmacology comparisons, for Wnt-pathway reporter and osteoblast differentiation studies, and as a tool reagent in assay development, ELISA/SPR/BLI standardization, and preclinical model work. It is offered at research grade with low endotoxin (typically less than 1 EU/mg, with an ultra-low-endotoxin option below 0.5 EU/mg) and can be produced at bulk milligram-to-gram scale to support screening campaigns and reproducible, batch-consistent studies. Sclerostin (SOST; UniProt Q9BQB4) is a secreted glycoprotein of the DAN/cerberus family, expressed predominantly by osteocytes embedded in bone. It is a key negative regulator of bone formation. Sclerostin binds the co-receptors LRP5 and LRP6 and antagonizes canonical Wnt/beta-catenin signaling, thereby restraining osteoblast activity, proliferation, and survival and dampening bone matrix deposition. It also modulates BMP signaling. Its activity is regulated mechanically and hormonally; mechanical loading and parathyroid hormone suppress SOST expression, permitting Wnt-driven bone accrual. Loss-of-function mutations cause the high-bone-mass disorders sclerosteosis and van Buchem disease, establishing sclerostin as a bone-mass brake. Because inhibiting sclerostin releases Wnt signaling and promotes bone formation, it is a validated therapeutic target in osteoporosis and a widely studied node in skeletal biology.
