Background
This is an unconjugated, non-therapeutic recombinant analog of the anti-CD3 x anti-BCMA bispecific antibody teclistamab, built as a human IgG4 (S228P/F234A/L235A)-lambda molecule that simultaneously engages CD3 epsilon on T cells and BCMA (TNFRSF17) on plasma cells. It is supplied for research use only and is not the clinical drug or intended for human or veterinary use. As a full-length T-cell engager format rather than a fragment, it is useful for studying redirected T-cell cytotoxicity, immune-synapse formation, and dual-target binding in reductionist systems. Typical research applications include positive/reference controls in BCMA- or CD3-directed bispecific assay development, benchmarking of in-house engagers, flow-cytometry and binding characterization, and preclinical in-vitro or ex-vivo models of T-cell redirection against BCMA-expressing targets. It is offered at research grade with low endotoxin (<1 EU/mg; ultra-low <0.5 EU/mg options), in bulk milligram-to-gram quantities suitable for assay scale-up and screening campaigns, giving labs a cost-effective, consistent reagent without originator-drug pricing or clinical-supply constraints. CD3 is a multi-subunit signaling complex (including the epsilon chain, UniProt P07766) associated with the T-cell receptor; clustering CD3 triggers the intracellular signaling that drives T-cell activation, proliferation, and cytotoxic effector function. BCMA (B-cell maturation antigen, TNFRSF17, UniProt Q02223) is a TNF-receptor-superfamily member whose expression is largely restricted to late-stage B cells and plasma cells; it binds APRIL and BAFF to support plasma-cell survival and is highly and selectively upregulated on malignant plasma cells in multiple myeloma. A bispecific engaging both antigens physically cross-links a cytotoxic T cell to a BCMA-positive target cell, forming an immune synapse that redirects T-cell killing independently of native TCR specificity. This restricted BCMA expression and the potency of CD3-driven activation make the pairing a widely studied model for T-cell-redirecting therapeutics.
