Background
This product is an unconjugated, non-therapeutic recombinant analog of favezelimab, an anti-LAG-3 (lymphocyte-activation gene 3) monoclonal antibody built on a human IgG4-kappa framework and directed against human LAG-3 (UniProt P18627). It is manufactured for research use only and is not the clinical drug, nor is it intended for human or veterinary use. As a research-grade reagent it is well suited to in-vitro and preclinical applications where a defined, sequence-consistent anti-LAG-3 binder is needed: as a positive control or benchmark in binding and blocking assays, as a tool for studying checkpoint biology in reconstituted or cell-based systems, for ADCC/ADC method development, and for characterizing LAG-3 expression and function on T cells. The IgG4 isotype limits effector engagement, consistent with a checkpoint-blocking design. Material is offered at low endotoxin (<1 EU/mg research grade; <0.5 EU/mg ultra-low-endotoxin options) and in bulk milligram-to-gram quantities to support in-vitro standards, assay-scale screening, and preclinical model work. LAG-3 (lymphocyte-activation gene 3; CD223) is a type I transmembrane immune-checkpoint receptor structurally related to CD4, encoded by the LAG3 gene (human UniProt P18627). It is expressed on activated CD4+ and CD8+ T cells, regulatory T cells, NK cells, and subsets of dendritic and B cells, and is a marker of T-cell exhaustion in chronic antigen exposure and tumors. Its best-characterized ligand is MHC class II, to which it binds with higher affinity than CD4; additional reported ligands include FGL1, galectin-3, and LSECtin. Engagement delivers an inhibitory signal that dampens T-cell proliferation, cytokine output, and effector function, and supports regulatory T-cell suppressive activity. Because LAG-3 frequently co-expresses with PD-1, it is a prominent co-inhibitory target, and blocking it can restore anti-tumor T-cell responses, motivating combination checkpoint strategies.
