Background
This product is an unconjugated, non-therapeutic recombinant analog of the anti-LAG3 antibody encelimab, provided strictly for research use only. It is engineered to reproduce the antigen-binding specificity of the originator molecule against human LAG3 (Lymphocyte Activation Gene 3, CD223) on a human IgG4-kappa framework, but it is not the clinical drug and is not intended for human or veterinary use. The recombinant format makes it a consistent, lot-controlled reagent for in-vitro and preclinical work: characterizing LAG3 expression and engagement on T cells, serving as a positive control or benchmark in binding and functional assays, supporting epitope and competition studies, and acting as a reference antibody in immune-checkpoint or immunomodulation research programs. It is supplied unconjugated so that users can label, immobilize, or reformat it as their workflow requires. Available in low-endotoxin research grade (<1 EU/mg) and ultra-low endotoxin (<0.5 EU/mg) formats and in bulk milligram-to-gram quantities, it is suited to assay development, screening, and preclinical model studies where reagent consistency and scalability matter. LAG3 (Lymphocyte Activation Gene 3, CD223; UniProt P18627) is a type I transmembrane immune-checkpoint receptor structurally related to CD4, with four extracellular Ig-like domains. It is expressed on activated CD4+ and CD8+ T cells, regulatory T cells, natural killer cells, B cells, and plasmacytoid dendritic cells. Its principal ligand is MHC class II, to which it binds with higher affinity than CD4; additional reported ligands include galectin-3, LSECtin, and fibrinogen-like protein 1 (FGL1). Engagement of LAG3 delivers inhibitory signals that dampen T-cell proliferation, cytokine production, and effector function, and it contributes to regulatory T-cell suppressive activity. Because LAG3 is frequently co-expressed with PD-1 on exhausted T cells in tumors and chronic infection, it is a major target for immune-checkpoint modulation and a focus of combination immunotherapy research.
