Background
This product is a research-grade biosimilar of amlenetug, an unconjugated non-therapeutic analog of the originator monoclonal antibody supplied for research use only. It is built around the same molecular target as the innovator drug, alpha-synuclein (SNCA), and is provided as a human IgG1, kappa immunoglobulin. The originator, amlenetug (also known as Lu AF82422), is an investigational antibody being developed as a potential disease-modifying treatment for alpha-synucleinopathies, most notably multiple system atrophy, where extracellular alpha-synuclein species are thought to drive pathological spreading. The originator recognizes an epitope in the C-terminal region of alpha-synuclein and engages monomeric, aggregated, and truncated forms of the protein. This biosimilar reproduces the binding specificity of that molecule so investigators can use it as a consistent, well-characterized tool reagent rather than as a clinical drug. It is manufactured to research-grade specifications with low endotoxin content and is available in bulk quantities to support reproducible in-vitro and functional studies. It is not a therapeutic product and is not intended for human or veterinary use.
Alpha-synuclein (SNCA, UniProt P37840) is a small, natively unfolded 140-amino-acid neuronal protein enriched at presynaptic terminals, where it is implicated in synaptic vesicle trafficking and neurotransmitter release. Its N-terminal region mediates lipid membrane binding, the central NAC domain drives aggregation, and the acidic C-terminus is a hub for interactions and post-translational modification. Under pathological conditions, alpha-synuclein misfolds and assembles into oligomers and amyloid fibrils that accumulate as Lewy bodies and glial cytoplasmic inclusions. These aggregates are the defining feature of synucleinopathies including Parkinson's disease, dementia with Lewy bodies, and multiple system atrophy. Extracellular and truncated species are thought to seed further aggregation and propagate pathology between cells in a prion-like manner. Antibodies recognizing alpha-synuclein, particularly its C-terminal region and aggregated conformers, are widely used to probe seeding, uptake, and clearance mechanisms.
