ICH5439 is a research-grade, unconjugated recombinant monoclonal antibody engineered as a non-therapeutic analog of cinpanemab, targeting human alpha-synuclein (SNCA). It is a human IgG1 lambda molecule expressed in a mammalian/recombinant system, built to mirror the variable-region specificity of the reference cinpanemab sequence so that researchers can study alpha-synuclein binding and aggregate biology without using clinical drug product. Cinpanemab is a human-derived antibody that preferentially recognises aggregated/pathological forms of alpha-synuclein, and was investigated as a passive-immunotherapy candidate for Parkinson disease. As a biosimilar analog this material is strictly for research use only (RUO) and is not intended for therapeutic, diagnostic or human/veterinary use. It is suited to functional and in-vitro work: alpha-synuclein binding and immunodepletion assays, aggregate/fibril capture, ELISA and immunoassay standardisation, seeding/aggregation and comparability studies, and mechanism-of-action characterisation. It is manufactured to rigorous quality standards with low endotoxin, supplied unconjugated and available in bulk (mg-g) quantities to support screening, assay development and larger functional studies. Alpha-synuclein (SNCA; also NACP/PARK1; UniProt P37840) is a small, natively unfolded 140-amino-acid presynaptic neuronal protein. It regulates synaptic vesicle trafficking and neurotransmitter release, acting as a monomer to promote vesicle priming and, in its multimeric membrane-bound state, as a chaperone assisting SNARE-complex assembly at presynaptic terminals; it also modulates dopamine neurotransmission via effects on the dopamine transporter. Its central (NAC) region drives a pathological conformational transition from soluble monomer to beta-sheet-rich oligomers and amyloid fibrils. These aggregates are the principal component of Lewy bodies and Lewy neurites and can propagate in a prion-like, cell-to-cell seeding manner. Misfolded and aggregated alpha-synuclein defines the synucleinopathies, including Parkinson disease, dementia with Lewy bodies and multiple system atrophy, making conformation- and aggregate-selective antibodies valuable tools for dissecting aggregation, spread and clearance.
