Background
This product is an unconjugated, non-therapeutic recombinant analog of the anti-CD137 (4-1BB/TNFRSF9) agonist antibody Exlinkibart, supplied as a human IgG1-kappa built around the TNFRSF9 target for research use only. It reproduces the binding specificity of the originator molecule so investigators can study agonistic engagement of the 4-1BB co-stimulatory receptor without sourcing clinical material. Typical applications include use as a defined positive control in binding and receptor-engagement assays, a reference reagent when benchmarking in-house 4-1BB agonists or bispecifics, and a tool for dissecting co-stimulatory signaling in T-cell and NK-cell systems. TNFRSF9, better known as 4-1BB or CD137 (UniProt Q07011), is a type I transmembrane member of the tumor necrosis factor receptor superfamily. It is an inducible co-stimulatory receptor expressed largely on activated CD8+ and CD4+ T cells, and also on NK cells, regulatory T cells, and dendritic cells. Its natural ligand is 4-1BBL (TNFSF9) presented on antigen-presenting cells. Ligand or agonist engagement triggers receptor clustering and recruitment of TRAF1 and TRAF2, driving NF-kB and MAPK (ERK, JNK, p38) signaling. Downstream effects include enhanced T-cell proliferation and survival, resistance to activation-induced cell death, increased cytokine production, memory CD8+ T-cell formation, and augmented cytotoxic and NK-mediated killing. These properties make 4-1BB a prominent co-stimulatory target for cancer immunotherapy and a widely studied node in T-cell activation biology.
