Background
This product is an unconjugated, non-therapeutic recombinant analog of rovalpituzumab, a human IgG1 antibody directed against Delta-like ligand 3 (DLL3; UniProt Q9NYJ7). It is supplied for research use only and is not the clinical drug, not the antibody-drug conjugate, and not intended for human or veterinary use. The molecule reproduces the DLL3-binding specificity of the originator in a bulk-scale, research-grade format, making it useful as a reference or benchmark reagent in target-validation and characterization work. Typical applications include use as a positive binding control, an isotype-matched comparator, and a building block for in-vitro assay development. Because DLL3 is a well-recognized tumor-associated antigen, the analog is frequently employed in ADC-development workflows, internalization and payload-delivery studies, and screening cascades where a defined anti-DLL3 binder is needed. It is offered at low-endotoxin research grade (<1 EU/mg) with an ultra-low option (<0.5 EU/mg), and in bulk milligram-to-gram quantities to support preclinical and assay-scale experiments.
Delta-like ligand 3 (DLL3) is an atypical Notch pathway ligand of the Delta/Serrate/Lag-2 family. Unlike canonical Notch ligands, DLL3 acts largely as an inhibitory (cis-acting) regulator, localizing predominantly to the Golgi and secretory vesicles where it restrains Notch signaling rather than trans-activating it on neighboring cells. In normal adult tissue DLL3 is expressed at very low levels and is largely intracellular, but in high-grade neuroendocrine tumors, notably small cell lung cancer and neuroendocrine prostate cancer, it is markedly upregulated and aberrantly trafficked to the cell surface. This tumor-restricted surface expression, driven downstream of the neuroendocrine transcription factor ASCL1, makes DLL3 an attractive tumor-associated antigen for targeted approaches including antibody-drug conjugates and T-cell engagers, and underpins its role as a differentiation and lineage marker in neuroendocrine biology.

Purification Method
