Background
This product is an unconjugated, non-therapeutic recombinant analog of lumretuzumab, an anti-HER3 (ERBB3) antibody, supplied strictly for research use only. It is a human IgG1/kappa molecule engineered to recognise human ERBB3 and reproduce the binding behaviour of the originator antibody, but it is not the clinical drug and is not intended for human or veterinary use. As a research reagent it is useful for probing ERBB3 biology and antibody function: positive/isotype-matched controls in binding assays, benchmarking of ERBB3-directed binders, ligand-blocking and receptor-internalisation studies, and as a building block for ADC or bispecific development and ADCC characterisation. It supports both in-vitro cell-based work and preclinical in-vivo experimental systems where a defined, well-characterised anti-ERBB3 tool is needed. The material is offered at research grade with low endotoxin (typically <1 EU/mg, with an ultra-low <0.5 EU/mg option) and is available in bulk milligram-to-gram quantities to support assay development, reagent standardisation, and scale-dependent screening. Formats are unconjugated to give users flexibility for downstream labelling or conjugation.
ERBB3 (HER3) is a member of the EGFR/HER receptor tyrosine kinase family. It contains an extracellular ligand-binding region, a single transmembrane domain, and a kinase-like domain that is catalytically impaired, so ERBB3 functions largely through heterodimerisation rather than as an active kinase on its own. Its principal ligands are the neuregulins (NRG1/heregulin), whose binding drives dimerisation with partners such as ERBB2 (HER2) and EGFR. The resulting heterodimers are potent activators of the PI3K-AKT survival and growth pathway, because ERBB3 carries multiple docking sites for the p85 subunit of PI3K. ERBB3 signalling contributes to normal epithelial and neural development, and aberrant ERBB3 activation is implicated in tumour proliferation, survival, and resistance to therapies targeting EGFR or HER2, making it a target of significant interest in oncology research.
