Background
This is a research-grade, unconjugated non-therapeutic recombinant analog of duligotuzumab, supplied for research use only (not for human or veterinary use). Duligotuzumab is notable as a dual-action antibody engineered to engage two receptors in the ErbB/HER family, and the analog is built around its receptor-binding activity for laboratory work. As a human IgG1/kappa protein produced recombinantly, it is useful as a defined, reproducible reagent for functional and preclinical studies: receptor-binding and neutralization assays, blocking of ligand-driven signaling, positive/isotype controls, epitope and competition experiments, and as a benchmarking standard alongside other anti-ErbB antibodies. The IgG1 backbone also makes it suitable for ADCC readouts and as a scaffold in antibody-drug-conjugate and bispecific development workflows. It is offered at research-grade low endotoxin (<1 EU/mg), with ultra-low endotoxin (<0.5 EU/mg) and bulk milligram-to-gram quantities available to support in-vitro assays and preclinical model standards. Being a non-therapeutic analog, it is intended purely for method development, characterization, and comparability rather than clinical application. ERBB3 (HER3, UniProt P21860) is a member of the EGFR/ErbB receptor tyrosine kinase family. It has an extracellular ligand-binding region, a single transmembrane domain, and a pseudokinase domain that is catalytically impaired, so ERBB3 cannot signal effectively on its own. Upon binding its ligands, the neuregulins (heregulins, NRG1/NRG2), ERBB3 heterodimerizes with partners, most potently ERBB2 (HER2) and also EGFR. Because its cytoplasmic tail carries multiple docking sites for the p85 subunit of PI3K, ERBB3-containing dimers are exceptionally strong activators of the PI3K/AKT survival and growth pathway, and also feed into MAPK signaling. ERBB3 up-regulation and neuregulin autocrine loops are recognized mechanisms of resistance to EGFR- and HER2-directed therapies, which is why ERBB3 and its EGFR/HER2 partners are prominent oncology targets.
