Background
This is a research-grade biosimilar of nezutatug: an unconjugated, non-therapeutic recombinant human IgG1-kappa monoclonal antibody engineered around the ERBB3 (HER3) target of the originator drug. It reproduces the antigen-binding specificity of the reference molecule and is intended strictly for research use, not for human or veterinary application. Supplied unconjugated, it suits functional and biochemical work such as ERBB3 binding and epitope characterisation, ligand (neuregulin/heregulin)-blocking and pathway-neutralisation assays, use as a benchmark or isotype-matched control alongside other anti-HER3 reagents, and as a starting scaffold for ADCC or antibody-drug-conjugate development. Material is offered at low endotoxin (research grade below 1 EU/mg; ultra-low grade below 0.5 EU/mg) and in bulk milligram-to-gram quantities, supporting in-vitro assay standardisation and preclinical study designs where reproducible, well-characterised antibody lots are needed. It is a research analog, distinct from the clinical product, and is provided for laboratory investigation only. ERBB3 (HER3) is a member of the EGFR/ERBB receptor tyrosine kinase family. Unlike its relatives, its intracellular kinase domain is catalytically impaired, so it signals mainly as a heterodimerisation partner. Binding of the ligands neuregulin-1 and neuregulin-2 (heregulins) to its extracellular region drives dimerisation, preferentially with ERBB2 (HER2), and trans-phosphorylation of ERBB3's cytoplasmic tail. Its multiple docking tyrosines recruit the p85 subunit of PI3K, making ERBB3 a uniquely potent activator of the PI3K-AKT survival and proliferation axis, with additional signalling through MAPK. ERBB3 upregulation and ligand-driven feedback are recurrent mechanisms of resistance to EGFR- and HER2-targeted therapies across breast, lung, gastric and other solid tumours, which is why blocking ligand binding or receptor dimerisation is an active therapeutic and research strategy.
