Background
This is a research-grade, unconjugated recombinant analog of elgemtumab (also written NRG1/HER3-directed anti-ERBB3 antibody), a fully human IgG1-kappa molecule that binds the extracellular region of human HER3 (ERBB3, UniProt P21860). It is supplied for research use only and is not the clinical therapeutic; it is not intended for human or veterinary use. The analog reproduces the target-binding specificity of the originator drug so that laboratories can use it as a defined tool for studying HER3-directed antibody activity: ligand-blocking and receptor-neutralisation assays, competition and epitope-binning experiments, ADCC/effector characterisation on HER3-positive cells, flow cytometry, and as a positive/reference control in biosimilar comparability or antibody-development workflows, including as an antibody-drug-conjugate scaffold. It is produced as bulk material at milligram-to-gram scale and offered in low-endotoxin formats (research grade less than 1 EU/mg; ultra-low less than 0.5 EU/mg) suitable for cell-based and preclinical model work. Being unconjugated, it can be paired with detection reagents or conjugation chemistries as the experiment requires.
ERBB3 (HER3, UniProt P21860) is one of four members of the ERBB/EGFR receptor tyrosine kinase family. Unlike its relatives, HER3 has an impaired intrinsic kinase domain and is considered catalytically weak; it signals chiefly by heterodimerising with a partner receptor, most importantly HER2 (ERBB2), and also EGFR and MET. Its principal ligands are the neuregulins (NRG1/heregulin), which bind the HER3 ectodomain and drive dimerisation. Once phosphorylated by its partner, HER3 provides multiple docking sites for the p85 subunit of PI3K, making it a potent activator of the PI3K/AKT survival and growth pathway. HER3 upregulation and NRG1 autocrine signalling are recognised mechanisms of resistance to HER2- and EGFR-targeted therapies, which is why blocking HER3 ligand binding or dimerisation is of research interest in oncology.
