Background
This is an unconjugated, non-therapeutic recombinant analog of livmoniplimab, supplied strictly for research use only. It is built as a human IgG4-kappa molecule directed against the GARP:latent-TGFβ1 complex, reproducing the target specificity of the originator drug so that laboratories can study GARP-mediated latent-TGFβ1 presentation and activation in controlled settings. It is not the clinical product and is not intended for human or veterinary use. Typical research applications include use as a binding and neutralisation reagent, an isotype-matched positive control in complex-directed assays, a benchmarking tool alongside other anti-GARP/TGFβ reagents, and a starting point for ADCC or antibody-engineering workflows. The material is offered at research grade with low endotoxin (<1 EU/mg research grade; <0.5 EU/mg ultra-low option) and is available in bulk milligram-to-gram quantities to support in-vitro assay development, preclinical characterisation, and formulation studies. GARP (glycoprotein A repetitions predominant), encoded by LRRC32, is a transmembrane leucine-rich-repeat protein that acts as a cell-surface docking receptor for latent TGFβ. It binds the latency-associated peptide (LAP) of pro-TGFβ1 (encoded by TGFB1) via disulfide bonds, tethering the latent complex to the membrane of regulatory T cells, activated platelets, and other cell types. This GARP:latent-TGFβ1 complex concentrates the cytokine at the cell surface and, together with integrins and mechanical force, promotes release of mature, active TGFβ1. Active TGFβ1 signals through TGFBR1/TGFBR2 and SMAD pathways to drive immunosuppression, Treg function, fibrosis, and tissue homeostasis. In tumours, GARP-dependent TGFβ1 activation contributes to an immunosuppressive microenvironment, making the surface-restricted GARP:latent-TGFβ1 complex an attractive, cell-context-specific target compared with systemic TGFβ neutralisation.
