Background
This is a research-grade, unconjugated recombinant analog of linavonkibart, a human IgG4-kappa antibody directed against transforming growth factor beta 1 (TGF-beta 1, TGFB1; UniProt P01137). It is supplied strictly for research use only and is not the clinical therapeutic; it is not intended for human or veterinary use. The molecule reproduces the antigen-binding specificity of the originator, allowing investigators to interrogate TGF-beta 1 biology in controlled experimental systems. Typical applications include in-vitro ligand neutralisation, receptor-binding and blocking assays, use as a positive isotype-matched control for IgG4 formats, and as a reference standard in ADCC/ADC and antibody-engineering workflows. It is offered in bulk milligram-to-gram quantities with low-endotoxin grades (research grade <1 EU/mg; ultra-low <0.5 EU/mg) to support cell-based and preclinical work where endotoxin would confound readouts. Because it is furnished as a defined, reproducible reagent, it is well suited to standardising assays across experiments and laboratories, benchmarking biosimilar candidates, and building assays around a well-characterised anti-TGF-beta 1 binder without reliance on scarce clinical material. Transforming growth factor beta 1 (TGFB1) is a secreted, disulphide-linked homodimeric cytokine and the prototypic member of the TGF-beta superfamily. It is produced as a latent complex in which the mature growth factor remains non-covalently associated with its latency-associated peptide; activation by integrins, proteases, or mechanical force liberates active ligand. Active TGF-beta 1 signals through the type II (TGFBR2) and type I (TGFBR1/ALK5) serine-threonine kinase receptors, driving SMAD2/3 phosphorylation and complex formation with SMAD4, alongside non-canonical MAPK and PI3K pathways. Biologically, TGF-beta 1 is a master regulator of cell proliferation, differentiation, extracellular-matrix deposition, epithelial-mesenchymal transition, and immune tolerance, including regulatory T-cell induction. Dysregulated TGF-beta 1 signalling contributes to tissue fibrosis, and to tumour immune evasion and metastasis, making it a target of intense therapeutic and mechanistic research interest.
