Background
This product is an unconjugated, non-therapeutic recombinant analog of the anti-CD47 antibody letaplimab (IBI-188), built as a human IgG4-kappa immunoglobulin directed against human CD47 (UniProt Q08722). It is manufactured for research use only and is not the clinical drug; it is not intended for human or veterinary use. The analog is useful as a well-defined reagent for interrogating the CD47-SIRPalpha "don't eat me" axis in vitro and in preclinical settings, for example as a positive control or benchmarking tool in binding, blocking, and macrophage phagocytosis assays, in antibody-dependent cellular phagocytosis (ADCP) and effector-function studies, and in ADC or bispecific development workflows where a defined CD47-targeting arm is needed. Because it shares the target specificity and IgG4 framework of the originator, it supports side-by-side comparison against novel candidates. It is supplied at research grade with low endotoxin (<1 EU/mg; ultra-low <0.5 EU/mg options) and is available in bulk milligram-to-gram quantities to support scale-up, assay standardization, and reproducibility across experiments. CD47 (Q08722), also called integrin-associated protein (IAP), is a widely expressed five-transmembrane glycoprotein with a single N-terminal IgV-like extracellular domain. Its principal counter-receptor is signal regulatory protein alpha (SIRPalpha) on macrophages and other myeloid cells; CD47-SIRPalpha engagement delivers an inhibitory "don't eat me" signal that suppresses phagocytosis, allowing cells to be recognized as self. CD47 also binds thrombospondin-1 and associates in cis with integrins, contributing to cell adhesion, migration, and nitric-oxide signaling. Many tumors upregulate CD47 to evade macrophage-mediated clearance, which is why blocking the CD47-SIRPalpha interaction is a validated strategy to restore phagocytosis and enhance antigen presentation. Because CD47 is also expressed on erythrocytes, on-target binding to red cells is a well-recognized consideration in experimental design.
