Background
ICH5249 is an unconjugated, non-therapeutic recombinant analog of itepekimab, an anti-IL-33 human IgG4-kappa antibody, supplied strictly for research use. It reproduces the antigen-binding specificity of the originator molecule, which was raised against human interleukin-33 (IL-33, UniProt O95760), so that laboratories can study IL-33 neutralization and IL-33/ST2 pathway biology without sourcing clinical material. The recombinant format gives lot-to-lot consistency and lets it serve as a defined reference or positive control in binding, blocking, and cell-based assays alongside other anti-IL-33 or anti-alarmin reagents. It is offered at low endotoxin (research grade <1 EU/mg; ultra-low <0.5 EU/mg options) and in bulk milligram-to-gram quantities to support screening cascades, assay standardization, and preclinical in-vitro work. As a human-target biosimilar analog it is intended for functional characterization, method development, and comparability studies, not for therapeutic, diagnostic, human, or veterinary use. Interleukin-33 (IL-33, UniProt O95760) is an IL-1 family cytokine and a member of the epithelial-derived "alarmins." It is constitutively expressed in the nucleus of barrier and structural cells, notably epithelial cells of the lung, gut and skin, and endothelial cells, where full-length IL-33 is bound to chromatin. On cell damage, necrosis, or stress it is released into the extracellular space, acting as a danger signal. Extracellular IL-33 signals through its receptor ST2 (IL1RL1) together with the co-receptor IL-1RAcP, driving MyD88-dependent NF-kB and MAPK activation. It potently activates type 2 immunity, stimulating ILC2s, Th2 cells, mast cells, basophils, eosinophils and regulatory T cells to produce IL-5 and IL-13. IL-33 bioactivity is modulated by proteolytic maturation and by oxidation, which limits its active window. It is strongly implicated in asthma, COPD and allergic and fibrotic disease.
