ICH5624 is an unconjugated, non-therapeutic recombinant analog of landogrozumab, a humanized anti-myostatin (MSTN/GDF8) antibody originally developed as an investigational agent for muscle-wasting conditions. This research-grade reagent is a full-length human IgG4-kappa immunoglobulin built around the same myostatin target, supplied for research use only and not intended for human or veterinary use. It is designed as a functional tool for laboratories studying myostatin biology, TGF-beta superfamily signaling, and muscle homeostasis. Typical applications include use as a neutralizing reagent in binding and ligand-blockade assays, as a positive control or benchmark antibody in comparative ADCC/effector-function or antibody-development workflows, and as a defined reference in preclinical in-vitro and cell-based models. Myostatin (MSTN), also known as growth/differentiation factor 8 (GDF8; UniProt O14793), is a secreted member of the TGF-beta superfamily that acts as a negative regulator of skeletal muscle mass. It is synthesized predominantly in skeletal muscle as a precursor that is proteolytically processed into an N-terminal propeptide and a C-terminal mature dimer; the propeptide keeps myostatin latent until activated. Mature myostatin signals by binding activin type II receptors (ActRIIB/ActRIIA), recruiting type I receptors (ALK4/ALK5), and driving SMAD2/3 phosphorylation, which restrains muscle-progenitor proliferation and protein accretion. Loss-of-function mutations produce marked muscle hypertrophy across species, including cattle, dogs, mice, and humans, establishing myostatin as a key brake on muscle growth and a focus of research into sarcopenia, cachexia, and muscular dystrophy.
