Background
This product is an unconjugated, non-therapeutic recombinant analog of domagrozumab, a fully human IgG1-kappa antibody directed against myostatin (MSTN/GDF-8). It reproduces the antigen-binding specificity of the originator molecule and is supplied for research use only; it is not the clinical drug and is not intended for human or veterinary use. The analog is useful as a defined, reproducible tool for interrogating myostatin biology in vitro and in preclinical systems: as a neutralising reagent to block mature myostatin signalling, as an isotype-matched positive control in binding, ELISA, SPR/BLI affinity, and cell-based reporter assays, and as a benchmarking standard when characterising other anti-myostatin binders or developing ADCs and engineered formats. It is offered at research grade with low endotoxin (<1 EU/mg; ultra-low <0.5 EU/mg options), in bulk milligram-to-gram quantities suitable for assay development, screening campaigns, and scale-up experiments. Because it is produced recombinantly to consistent specifications, it supports lot-to-lot reproducibility in comparative and mechanistic studies of the myostatin/activin-SMAD axis.
Myostatin (MSTN), also known as growth differentiation factor 8 (GDF-8), is a member of the TGF-beta superfamily and a dominant negative regulator of skeletal muscle mass. It is synthesised as a precursor that is proteolytically processed into a latency-associated propeptide and a C-terminal mature dimer; the propeptide keeps circulating myostatin latent until further cleavage releases active ligand. Mature myostatin binds the activin type II receptors (ActRIIB, and ActRIIA) with a type I receptor (ALK4/ALK5), driving SMAD2/3 phosphorylation and transcriptional programs that suppress myoblast proliferation and differentiation and promote proteolysis. Loss-of-function mutations produce marked muscle hypertrophy across species, making myostatin a long-standing target for muscle-wasting, sarcopenia, and dystrophy indications. Its signalling overlaps with activins and GDF11, so target specificity is an important experimental consideration.
