Background
ICH5454 is a research-grade biosimilar of crenezumab, supplied as an unconjugated non-therapeutic analog of the originator antibody for research use only. It reproduces the target specificity and human IgG4-kappa framework of crenezumab but is intended for laboratory and functional work, not clinical or therapeutic use. Crenezumab is a humanized monoclonal antibody directed against amyloid-beta (Abeta), the peptide derived by proteolytic processing of amyloid precursor protein (APP, UniProt P05067). It was developed as an Alzheimer's disease candidate and is notable for its engineered IgG4 backbone, chosen to limit engagement of Fc-gamma receptors and thereby reduce the risk of microglial over-activation, neuroinflammation, and vasogenic edema seen with some other anti-Abeta antibodies. This biosimilar is well suited as a reference or comparator reagent in binding, aggregation, and neutralization studies of Abeta species. It is offered at research and in-vivo grades with low endotoxin and in bulk quantities. Note that the card lists the target as APP (P05067); the functional epitope is the Abeta peptide region rather than full-length APP.
Amyloid precursor protein (APP, UniProt P05067) is a ubiquitously expressed transmembrane protein whose sequential cleavage by beta- and gamma-secretases liberates the amyloid-beta (Abeta) peptide. Abeta monomers can self-associate into soluble oligomers and insoluble fibrils that accumulate as extracellular plaques, a defining pathological hallmark of Alzheimer's disease. Soluble Abeta oligomers are widely regarded as the most synaptotoxic species, disrupting synaptic function and driving neurodegeneration. Crenezumab binds an epitope in the mid-region of Abeta (approximately residues 13-24) and preferentially recognizes aggregated oligomeric and fibrillar assemblies with high affinity while binding monomeric Abeta weakly. Functionally it can block Abeta aggregation, promote disaggregation, and, through its IgG4 backbone, favor microglial clearance of aggregates with minimized Fc-gamma receptor activation.
