Background
Clone R4-6A2 is a rat IgG1 kappa monoclonal antibody raised against mouse interferon-gamma (IFN-gamma). It is one of the most widely used and long-established anti-mouse IFN-gamma clones, recognised for its ability to both capture and neutralise the cytokine, which is why it appears routinely as a capture reagent in ELISA and ELISPOT assays and as a neutralising reagent in functional studies. This ichor.bio format is validated for ELISA capture, ELISPOT, neutralization, and frozen-section immunohistochemistry. It is manufactured to a low-endotoxin specification (research-grade <1 EU/mg; ultra-low <0.5 EU/mg) and supplied in-vivo-ready and in bulk milligram-to-gram quantities, making it well suited to in-vivo neutralisation experiments and to functional in-vitro and ex-vivo work where endotoxin contamination would confound cytokine and immune-cell readouts. The rat IgG1 backbone is a comparatively low-effector isotype, favouring cytokine neutralisation over Fc-mediated effector engagement. Interferon-gamma (IFN-gamma, encoded by Ifng; UniProt P01580) is the sole type II interferon and a central pro-inflammatory, immunoregulatory cytokine. It is produced chiefly by activated CD4+ Th1 cells, CD8+ cytotoxic T cells, natural killer and NKT cells, and to a lesser extent by innate lymphoid and myeloid populations. The biologically active form is a homodimer that signals through the heterodimeric IFN-gamma receptor (IFNGR1/IFNGR2) via the JAK1/JAK2-STAT1 pathway, driving transcription of interferon-stimulated genes. Functionally, IFN-gamma activates macrophages, upregulates MHC class I and II antigen presentation, promotes Th1 polarisation, enhances antimicrobial and antitumour immunity, and contributes to tissue inflammation and immunopathology. Because of this broad role, it is a key readout and target across infection, autoimmunity, and tumour immunology research.
