Background
This product is clone H22, an Armenian hamster IgG monoclonal antibody raised against mouse interferon gamma (IFN gamma, UniProt P01580). H22 is a well-characterized neutralizing clone widely used to block IFN gamma bioactivity in mouse models, making it suited to in-vivo and functional studies of type II interferon signaling. Because it is of hamster origin and directed against a mouse cytokine, it is appropriate for systemic administration in mouse experiments without the confound of an anti-mouse-IgG response seen with some rat clones. Validated applications include neutralization, ELISA, immunoprecipitation, Western blot, and immunofluorescence. ichor.bio supplies this antibody as a low-endotoxin (research grade less than 1 EU/mg; ultra-low option less than 0.5 EU/mg), in-vivo-ready preparation available in bulk milligram-to-gram quantities. It is offered for research use only (RUO). The low endotoxin load reduces the risk of confounding innate immune activation, which is important when interpreting cytokine-blockade phenotypes in immunology, infection, autoimmunity, and tumor studies. Researchers commonly search for this reagent by the clone designation H22. Interferon gamma (IFN gamma) is the sole type II interferon and a central regulator of cell-mediated immunity. In mice it is encoded by Ifng and produced predominantly by activated T helper 1 (Th1) CD4 cells, CD8 cytotoxic T cells, natural killer cells, and NKT cells. It signals through a heterodimeric receptor (IFNGR1/IFNGR2) that activates the JAK1/JAK2-STAT1 pathway, driving transcription of interferon-stimulated genes. Functionally, IFN gamma activates macrophages to enhance microbicidal activity, upregulates MHC class I and class II antigen presentation, promotes Th1 differentiation while suppressing Th2 and Th17 responses, and contributes to antitumor immunity. Dysregulated IFN gamma is implicated in autoimmune and inflammatory disease, hemophagocytic lymphohistiocytosis, and both protective and pathological responses during infection. Neutralizing IFN gamma in vivo is a common strategy to dissect its role across these contexts.
