Background
This product is an unconjugated, non-therapeutic recombinant analog of sofituzumab, supplied for research use only and built around the MUC16 (CA125) target. It reproduces the antigen-binding specificity associated with the originator anti-MUC16 antibody without any drug payload, radiolabel, or conjugation, making it a clean reagent for target-engagement and mechanism studies rather than a clinical material. It is not the clinical drug and is not intended for human or veterinary use. Researchers use it as a defined binding tool to interrogate MUC16 biology, as a matched isotype-format control in binding and effector assays, and as a starting reagent for antibody-drug-conjugate and effector-function development workflows where a well-characterized anti-MUC16 binder is needed. It is offered as a human IgG1 kappa format molecule. Available at research grade with low endotoxin (<1 EU/mg), with ultra-low endotoxin (<0.5 EU/mg) and bulk milligram-to-gram quantities suitable for in-vitro assays, preclinical characterization, and assay standardization. Lot-to-lot consistency and purity support reproducible use across binding, neutralization, and screening applications.
MUC16 (UniProt Q8WXI7), also known as CA125, is a very large, heavily O-glycosylated transmembrane mucin. It comprises an extensive N-terminal region, a central tandem-repeat domain bearing SEA modules, and a short membrane-proximal ectodomain, transmembrane segment, and cytoplasmic tail. Proteolytic cleavage releases much of the extracellular portion as shed CA125, the widely used serum biomarker for epithelial ovarian cancer, while a retained membrane-proximal fragment stays cell-associated. MUC16 contributes to a protective, hydrated mucosal barrier at epithelial surfaces and is markedly overexpressed in ovarian and some pancreatic and other carcinomas. It engages binding partners such as mesothelin, supporting cell adhesion and peritoneal metastasis, and its cytoplasmic tail can modulate intracellular signaling. These features make the retained, cell-bound portion of MUC16 an attractive tumor-associated antigen for targeted approaches.
