Background
This product is an unconjugated, non-therapeutic research-grade analog of the originator antibody Abagovomab, supplied for research use only (RUO) and not for clinical or diagnostic use. Abagovomab is notable as an anti-idiotypic (Ab2) monoclonal antibody: rather than binding the tumor antigen CA 125 (MUC16) directly, it was raised against the murine anti-CA 125 antibody OC125 so that its variable region structurally mimics the CA 125 epitope. In the originator setting it was designed to act as a surrogate antigen/immunogen, provoking an anti-anti-idiotypic (Ab3) response able to recognize CA 125-expressing ovarian tumor cells. The originator was developed as maintenance immunotherapy in ovarian cancer but was discontinued after the Phase III MIMOSA trial did not meet its recurrence-free or overall survival endpoints. As a biosimilar analog, this reagent (human IgG1/kappa framework) is intended for functional and in-vitro studies of idiotype-based immune mechanisms and CA 125 biology. Because of its unusual anti-idiotypic mechanism, investigators should confirm the reagent's binding behavior matches their assay design before use. CA 125 is the classic serum biomarker epitope carried on Mucin-16 (MUC16; UniProt Q8WXI7), an extremely large, heavily O-glycosylated transmembrane mucin. MUC16 is over-expressed by more than 80% of epithelial ovarian cancers and is shed into circulation, making CA 125 the most widely used marker for monitoring ovarian cancer burden and treatment response. At the cell surface the mucin contributes to a protective glycocalyx, modulates cell adhesion and immune evasion, and can engage partners such as mesothelin and galectins. Importantly, Abagovomab does not recognize MUC16/CA 125 itself; it is an anti-idiotypic antibody whose paratope mimics the CA 125 epitope defined by the reference antibody OC125. The relevant biology for this reagent is therefore idiotype/anti-idiotype network immunology as applied to CA 125, rather than direct antigen engagement of the mucin.
