Background
ICH5093 is a recombinant, research-grade biosimilar analog of pinatuzumab, an anti-CD22 (Siglec-2) antibody. It is supplied as an unconjugated, non-therapeutic human IgG1-kappa protein that reproduces the antigen-binding specificity of the originator molecule while omitting the auristatin payload and cleavable linker used in the parent antibody-drug conjugate. This makes it well suited as a target-specific tool for in-vitro and preclinical research rather than for clinical use. Typical applications include use as a defined positive/reference binder in CD22 assays, as an isotype-matched or specificity control alongside other B-cell reagents, as a capture or detection reagent for CD22-expressing cells, and as a naked-antibody backbone for evaluating internalization, ADCC, or ADC and payload-conjugation development workflows. The material is offered at research-grade purity with low endotoxin (research grade less than 1 EU/mg; ultra-low less than 0.5 EU/mg options) and is available in bulk milligram-to-gram quantities to support assay development, scale-up, and reproducibility. It is for research use only (RUO) and is not the clinical drug; it is not intended for human or veterinary use. CD22, also called Siglec-2 (UniProt P20273), is a type I transmembrane glycoprotein of the sialic-acid-binding immunoglobulin-like lectin (Siglec) family and a defining B-lineage marker. Its expression is largely restricted to B cells, appearing during maturation and present on most mature B cells but generally lost on plasma cells. The extracellular region comprises seven Ig-like domains, with the N-terminal domain mediating recognition of alpha-2,6-linked sialic acid on both cis (same-cell) and trans ligands. The cytoplasmic tail carries immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that, upon phosphorylation, recruit the phosphatase SHP-1 to dampen B-cell receptor signaling and calcium flux, positioning CD22 as a negative regulator of B-cell activation and a contributor to tolerance. Antibodies bound to CD22 are rapidly internalized, which underlies its longstanding use as a delivery target in B-cell malignancies such as non-Hodgkin lymphoma and leukemia.
